The Time Burst is the signature attack. The room darkens, then explodes after a few seconds for 999 damage, which is essentially a guaranteed kill or a spent Death Defiance unless you reach the lit safe number on the clock. He uses both a small version and a big centered version, and either one ends a careless run instantly.
Bring real burst damage, save your Death Defiances for the Time Bursts, and never plan around time boons, because his passive turns them off.
The most important rule is the safe zone. When the second-phase arena darkens for a Time Burst, a single clock number lights up as the safe spot. Sprint there immediately rather than dodging in place, because the blast covers everything else and deals 999 damage. Treat that light as the only thing that matters for those few seconds.
Manage his healers and shields too. The Tempus adds in phase two fire a green beam that heals Chronos for 50 each, so kill them quickly or his bar stops moving. In phase one, destroy the Time Banners before their pulse reaches him, since each one stacks a Shield on him and his minions. A Hermes 
speed boon helps you cross the arena to both the safe zones and the adds in time.
Front-load your damage. Chronos regains all of his health between phases, so there is no value in slow chip damage; you want to delete each phase bar fast and keep enough Death Defiance in reserve for the second-phase Time Bursts. In practice, a high-burst build with a dash defense beats him far more reliably than a tanky grind.
His Time Immunity passive is the planning constraint: every time-manipulation trick you have built around stops working, so lean on raw damage and defense instead.
