Each main stage has a unique boss that spawns at 25:00. Defeat the boss to unlock Hyper Mode for that stage. The five normal stages are Mad Forest, Inlaid Library, Dairy Plant, Gallo Tower, and Cappella Magna.

Beyond the standard hyper bosses, several base-game stages have additional encounters. Cappella Magna has The Ender as a 30:00 endgame boss when the Yellow Sign is collected. The Bone Zone has Sketamari, a katamari-style rolling skeletal mass that absorbs other enemies.

Reapers are end-of-stage enemies that spawn at 30:00 to terminate the run. The standard Reaper has variants in different stages: Drowner (Gallo Tower), Stalker (Dairy Plant), Maddener (Holy Forbidden), and Trickster (Cappella Magna). All are normally invulnerable.
The Reaper family is one of the most iconic mechanics in Vampire Survivors. The standard Reaper spawns at the 30-minute mark on every stage to forcibly end the run. Each variant has a unique theme tied to its home stage.

Cappella Magna features Reaper Trainees as standard wave enemies. They come in seven colour variants: Yellow, Blue, Gray, Green, Purple, Red, and Elite. Each has different stats. Defeating a non-Red Reaper Trainee with an uppercut motion triggers the Toastie character unlock. See the

The Director is the canonical final boss of Vampire Survivors, encountered on the Eudaimonia Machine special stage. The fight has four phases plus a victory phase. Defeating The Director unlocks the Greatest Jubilee weapon and the ending credits.

The Director cannot be damaged directly. Each of her four phases is cleared by destroying the orbital fragments she protects: seven Atlantean masks in Phase 2, five skulls and two eyes in Phase 3, and coin/gem clusters in Phase 4. Mt.Moonspell adds Orochimario as the 25:00 hyper boss. The stage also contains the Kappa enemy in the northwestern lake area, which unlocks Gav'Et-Oni after 6,000 kills.

Orochimario is the hyper boss of Mt.Moonspell, spawning at 25:00. The boss is based on the Yamata no Orochi yokai and has eight heads. Each head can be defeated individually, with the first head having double health (1,000 stars) and the total health being 5,000 stars.
Mt.Moonspell also features the
Kappa yokai in the northwestern lake area. The Kappa is bestiary entry #183 and serves as the unlock condition for Gav'Et-Oni: defeat 6,000 of them to add the character to the roster.

Lake Foscari contains the standard hyper boss. Abyss Foscari adds Je-Ne-Viv as a special boss tied to character unlocks, plus the Blinder event enemy.
The Tides of the Foscari DLC introduces two stages: Lake Foscari (the surface map with the magical academy) and Abyss Foscari (the dark subterranean realm). Both contain unique bosses.

The Foscari DLC also introduces the SpellString, SpellStream, and SpellStrike weapons used to interact with the colored emerald gates in the academy. Lake Foscari does not have a 25:00 hyper boss in the traditional sense, instead using character-specific gate interactions for progression.
Polus Replica is the Among Us crossover stage. Suspicious Eyes is the 25:00 hyper boss. Megalo Impostor Rina is unlocked by defeating 100,000 enemies in a single Impostor Rina run.

Neo Galuga is the Contra crossover DLC stage. Big Fuzz is the 25:00 hyper boss. Defeating Big Fuzz unlocks Colonel Bahamut as a playable character.
The Operation Guns DLC adds Neo Galuga (and the bonus Hectic Highway stage). The boss roster is themed after Contra and similar run-and-gun titles.

Death is the major endgame boss of Ode to Castlevania, accessed via the Throne Room with Richter Belmont. The fight occurs after a dialogue between Richter and Dracula in which The Director helps put Vlad Tepes Dracula into a coffin (unlocking him as a playable character).
