Hive Knight is the fight Team Cherry added specifically to give the Hive zone a boss reason to exist. Per the wiki, "Hive Knight is a boss in Hollow Knight, introduced in Lifeblood. Defeating him grants a new Charm, Hiveblood." Before the Lifeblood update, the Hive ended at Queen Vespa's lifeless body with no fight; the patch added the boss and the Charm reward together.

Hive Knight sits at the eastern end of the Hive. Per the wiki, "Hive Knight can be fought in the furthest room in the Hive where the giant lifeless body of the queen lies. He ambushes the Knight as they enter the place and battles them ferociously." The arena triggers on entry; backing out exits the room and resets the fight.
According to the wiki, the Hive itself is gated behind the Tram in Kingdom's Edge. Players need Tram Pass + Crystal Heart or equivalent traversal to reach the boss room. The full Hive corridor requires beating Hive Soldiers, Hive Guardians, and Husk Hives to clear the path to the queen's chamber.

Per the wiki, "Hive Knight is a light boss. As such, the Knight has more opportunities to move Hive Knight than many other bosses. Steady Body is very strong against Hive Knight because it allows the Knight to maintain pressure. Quick Slash is also strong against Hive Knight, provided the Knight does only upward slashes (otherwise Hive Knight is knocked away and the Knight constantly has to chase)." In actual play, up-slashes pin him against the floor for stagger races.
The standard Hive Knight loadout uses Shade Cloak invincibility to dash through Surprise Slash and Lunge. Per the wiki, "it is highly recommended to obtain Shade Cloak before the fight, as this makes it possible to dash through Hive Knight's Surprise Slash and Lunge attacks and makes handling the Swarm Release attack safer."

There are plenty of no-Charm healing opportunities too: when out of range of a Surprise Slash or Lunge attack, also if dodged early; during Honey Spikes (when in a safe location); at the beginning of Swarm Release; and during Hive Knight's stagger." Heal early in Swarm Release, not late.
The wiki specifies the spell pick. "The Vengeful Spirit / Shade Soul Spell is strong against Hive Knight due to its versatility." Combined with Shaman Stone, spells handle Hivelings during Swarm Release and finish Hive Knight from cross-arena range during Honey Spikes.
The wiki flags Cyclone Slash as a high-DPS Nail Art option. "The Cyclone Slash Nail Art is very strong against Hive Knight due to the Knight being able to move horizontally while repeatedly pressing the attack button." In real runs this is the cleanest stagger-rush; one Cyclone covers half the stagger threshold solo.
Hive Knight drops the Hiveblood Charm. Per the wiki, the Charm restores the last Mask taken if the Knight avoids damage for 10 seconds. It does not heal other Masks; only the most recent. In practice this is a Steel Soul (no-Lifeblood) staple and a clutch Charm for tight rooms in Pantheons.
According to the wiki, Hive Knight returns in the Godmaster content as the opening boss of the Pantheon of the Sage. "Pantheon of the Sage, 1st boss. Health: Attuned difficulty 850. Ascended difficulty 1300." The Godhome arena uses the same layout as the Hive but "starts without the 7 Hivelings descending. Can use all attacks from the start on Ascended difficulty or above."
The wiki notes the Hall of Gods tag. "I guard the heart of the hive for my Queen. Watchful god of duty." This frames Hive Knight as the archetypal loyal-protector boss in Hallownest's pantheon, similar in role to Dryya among the Five Great Knights.
